Haptoglobin Phenotypes and Levels in Inflammatory Arthritis Patients

Abstract
Haptoglobin (Hp) phenotypes and levels were studied in the inflammatory arthritis patients and the healthy control group.
Serum samples were obtained from 152 inflammatory arthritis patients and 251 unrelated healthy blood donors, living in Seoul, Korea. Typing of Hp was performed by horizontal electrophoresis in 5 % polyacrylamide gel using a discontinuous buffer system. Hp concentration was measured by a single radial immunodiffusion.
The healthy controls were observed three phenotypes: Hp 1?1, Hp 2?1 and Hp 2?2 were 25, 103 and 123, respectively. Allele frequencies were estimated Hp¢¥ 1 at 0.305 and Hp¢¥ 2 at 0.695. In patients, Hp 1?1, Hp 2?1, Hp 2?2 and Hp 0 were 20, 61, 70 and 1 respectively. Allele frequencies were estimated Hp* 1 at 0.334 and Hp* 2 at 0.666. There were no significant differences in gene or phenotype frequencies between the patients and the controls.
Hp levels were significantly different among genotypes. Serum Hp concentration in the patients were 303.6 ¡¾ 225.6 mg/100 ml (N=19) in the Hp 1?1, 516.4 ¡¾ 220.9 mg/100 ml (N = 60) in the Hp 2?1, and 478.4 ¡¾ 246.8 mg/100 ml (N=70) in Hp 2-2; the healthy controls were 119.9 ¡¾ 49.2 mg/100 m1 (N =25) in the Hp 1?1, 213.3 ¡¾ 95.4 mg/100ml (N=103) in the Hp 2?1, and 14&.1 ¡¾88.4 mg/100 ml (N= 123) in the Hp 2?2. The amount of serum haptoglobin markedly increase in the inflammatory arthritis patients compare with the healthy controls(p<0.001).
